摘要

摘 要:为探究槲皮素对人软骨肉瘤细胞迁移、侵袭和上皮间质转化(EMT)的影响及对Janus激酶2/信号转导和转录启动因子3(JAK2/STAT3)信号通路的调控作用,体外培养人软骨肉瘤SW-1353细胞,用不同浓度槲皮素干预SW-1353细胞24 h,细胞计数试剂盒(CCK-8)法筛选出最适浓度(50.0 μmol/L)槲皮素进行后续试验。将细胞分为对照组(不做干预)、槲皮素组(50.0 μmol/L槲皮素)、阳性药物组(6.0 μmol/L阿霉素)、抑制剂组(50.0 μmol/L槲皮素+50.0 μmol/L JAK2/STAT3通路抑制剂AG490)和激活剂组(50.0 μmol/L槲皮素+0.5 μmol/L JAK2/STAT3通路激活剂colivelin),干预24 h后,用倒置显微镜、Transwell小室及Western blot法对细胞形态、迁移、侵袭及JAK2/STAT3通路和EMT相关蛋白的表达水平进行分析。试验结果显示:CCK-8法确定槲皮素最适作用浓度为50.0 μmol/L;与对照组比较,槲皮素组和阳性药物组细胞生长受到抑制,细胞迁移数、侵袭数、N-钙黏蛋白(N-cadherin)、波形蛋白(Vimentin)、纤连蛋白(FN)、p-JAK2和p-STAT3表达水平降低(P<0.05),E-钙黏蛋白(E-cadherin)表达水平升高(P<0.05);与槲皮素组相比,抑制剂组上述指标变化趋势更为显著(P<0.05),激活剂组则扭转了这些指标的变化(P<0.05)。槲皮素可抑制人软骨肉瘤SW-1353细胞的生长、迁移、侵袭和EMT进程,其作用机制可能与阻滞JAK2/STAT3信号通路相关。

Abstract

Abstract: To investigate the effects of quercetin on human chondrosarcoma cell migration, invasion and epithelial interstitial transformation (EMT) and the regulatory role of Janus kinase 2/signal transduction and transcription promoter 3 (JAK2/STAT3) signaling pathway, human chondrosarcoma SW-1353 cells were cultured in vitro and treated with quercetin of different concentrations for 24 h. The optimal concentration of quercetin (50.0 μmol/L) was selected by cell counting kit (CCK-8) for subsequent experiments. The cells were divided into control group with no intervention, quercetin group with 50.0 μmol/L quercetin, positive control group with 6.0 μmol/L adriamycin, inhibitor group involving 50.0 μmol/L quercetin +50.0 μmol/L JAK2/STAT3 pathway inhibitor AG490, and activator group involving 50.0 μmol/L quercetin +0.5 μmol/L JAK2/STAT3 pathway activator colivelin. After 24 h of intervention, cell morphology, migration, invasion, and expression levels of JAK2/STAT3 pathway and EMT-related proteins were analyzed by inverted microscope, Transwell chamber and Western blot. The experimental results showed that the optimum concentration of quercetin was determined by CCK-8 method to be 50.0 μmol/L. Compared with the control group, the growth of quercetin group were positive control group were inhibited, the cell migration number, invasion number, expression levels of N-cadherin, Vimentin, fibronectin (FN), p-JAK2 and p-STAT3 in quercetin group and positive control group decreased (P<0.05), E-cadherin expression level increased (P<0.05); compared with quercetin group, the change trend of the above indexes in inhibitor group was more significant (P<0.05), while the change of these indexes in activator group was reversed (P<0.05). Quercetin can inhibit the growth, migration, invasion and EMT progression of human chondrosarcoma SW-1353 cells, possibly by blocking the JAK2/STAT3 signaling pathway.
