Application of DNA Detection by Surface Enhanced Raman Scattering

Surface enhanced Raman scattering (SERS) is an optical spectroscopy that has been widely used in biomedical analysis. It is capable of extremely sensitive biomolecule detection and multiplexed analysis. This perspective highlights where SERS has been used to detect DNA, proteins, and where in vivo and in vitro analysis has been successfully utilized. SERS has becoming an alternative novel technique for biomolecule detection and clinical diagnostics.

Application of optical coherence tomography in endoscope

Optical coherence tomography (OCT) is a new imaging technique after X-ray imaging, magnetic resonance imaging, ultrasonic imaging and so on, and its characteristic of can be optical fiber make it easy to combine with medical electronic endoscope. OCT endoscope technology can realize the high rate, high resolution, non-destructive and real-time imaging of human internal organs. The classification and basic principles of OCT as well as some important parameters of OCT technology including the probing depth and axial resolution are mainly introduced. Some recent research achievements and development of OCT endoscope scanning probes are discussed. The influences of light source on OCT endoscope are mostly analyzed. The main application of OCT endoscope is summarized.

Mesoporous Carbon Nanospheres: Synthesis and Application in Drug Delivery

The advances in functional nanomaterials offer great opportunities for the development of nanomedicine. In the last decade we have seen great progress in the synthesis and application of mesoporous carbon nanomaterials. As a new inorganic material, mesoporous carbon nanomaterials integrate the advantages of a mesoporous structure and carbonaceous composition, and have shown superior properties  compared with traditional mesoporous silica and other carbonbased materials, such as carbon nanotube, graphene, and fullerene. Mesoporous carbon nanomaterials have shown great promise in the fields including drug delivery and controlled release, photothermal therapy, synergistic therapy, cellstaining, catalysis, biosensor, and macromolecular separation. This review summarizes the primary techniques for preparing and modifying mesoporous carbon nanomaterials, with special emphasis on the applications of these materials in drug delivery and ondemand photothermaltriggered drug release. Finally, we present the challenges and opportunities in the development and clinical translations of this kind of nanomaterials.

Study Progress of the Characterization and Biological Function of GYF Domain Protein
GYF domains are small, conservative, versatile adaptor domains that recognize prolinerich sequences (PRS). GYF domain proteins play important roles in protein protein interaction, translational initiation, mRNA splicing, surveillance and translation suppression, ubiquitin ligation, signal transduction and immune protease regulation, et al. The study progress of GYF domain protein on aspects of domain characters, interaction with target proteins as well as its function were reviewed in the paper.

A Fast Photoacoustic Microscope Imaging System Based on Linear Motor  ContinuousStepping Motion
A system for twodimensional photoacoustic imaging with a continuousstepping scan of a linear motor is reported. The system is in the form of a bowtype curve during motion, in which the linear motor moves continuously in the Xaxis direction, Yaxis direction to have a step by step movement, capture card in the Xaxis motor movement in the process of continuous acquisition. The imaging system is larger than the previous method of scanning the galvanometer, and can reach the biological tissue photoacoustic imaging in the centimeter scale range. Compared with the previous stepping motor, the scanning speed can be improved obviously. At the same time, Driving the light and ultrasonic transducers without moving  the motor can make the sample scanning more flexible.In addition, the use of embedded carbon wire simulation samples and in vivo mouse ear blood vessels to verify the imaging capabilities of the system. The experimental results show that this fast photoelectrical twodimensional microscopic imaging system can be used to achieve highresolution imaging in vivo, and is expected to be a noninvasive, realtime photoacoustic microscope for biomedicine.

Terahertz Timedomain Spectroscopy of Melatonin
Terahertz timedomain spectroscopy is a new nondestructive detection technique, which uses the property of low power of terahertz wave, and the characteristics of strong dispersion and absorption of the vibrational transitions and rotations of most biomolecules are shown in this frequency band. These make it possible,to have nondestructive detection and the study of biological activities. In this work, transmission and reflection of terahertz timedomain systems were respectively used to carry out testing melatonin tablets with different mass ratio, and analysis of its spectral properties in terahertz band were also given. It could be concluded  that it had characteristic absorption peak at 0.29, 0.50, 0.70, 0.91, 1.20, 2.17 and 2.55 THz. The intensity of the frequency spectrum has a linear relation with the concentration of the sample. In addition, Gaussian 09 and Gaussian VIEW software were used to perform simulation, and the characteristic absorption peaks of melatonin in 0.46, 0.91, 1.15, 2.01, 2.23 and 2.61 THz were acquired, which provided a strong support for experiment results. All these works are the basis and reference for the detection and identification of biochemical samples such as melatonin.

Largefield Photoacoustic Microscopy with Rapid Scanning Based on Linear Motor
Photoacoustic imaging, which was developed in recent years, is a new and noninvasive medical imaging technology. The imaging technique uses laser pulses to generate photoacoustic signal, and transmit the tissue information with ultrasonic, and reconstruct the image of tissue structure and function information with the corresponding reconstruction algorithm. It combines the characteristic of optical imaging and ultrason imaging, provides high resolution and high contrast tomographic images of the deep tissue and has a wide range of applications in biomedical clinical diagnosis as well as in vivo imaging. Currently, there are two scanning modes for photoacoustic microscopy:photoacoustic microscopy system based on twodimensional scanning galvanometer and photoacoustic microscopy system based on stepping motor. In this work, considering the limitations of the low speed of stepping motor based photoacoustic microscopy (0.001 frame/s) and small scanning scope of scanning galvanometer based photoacoustic microscopy (<1 mm2), we developed an largefield photoacoustic microscopy with rapid scanning based on linear motor. The scanning speed of this technique is 200 mm/s。The technique is based on a linebyline method to collect the photoacosutic signal, which is 800 times faster than the stepmotor based photoacoustic microscopy with pointbypoint gathering of photoacosutic signal. The scanning speed of this system is 0.8 frames per second in the field of 10 mm×10 mm. The maximum scanrange can reach 50 mm×50 mm. The development of the  large field rapid photoacoustic microimaging system will provide new imaging tools for biomedical science.

A Study on Cisplatin Targeting Cx43 Expression in Cell Cycle of Esophageal Cancer
Objective:To study the target effect of cisplatin on treating esophageal squamous cell carcinomas (ESCC). 〖WTHZ〗Methods:〖WTBZ〗In this study, the influence of cisplatin on cell cycle progression of esophageal cancer and connexin 43 (Cx43) expression in cancer cell cycle were detected by bivariate flow cytometric analysis. Results:he effect of cisplatin on cell cycle of ESCC was mainly due to DNA replication of S phase cells, arrest of cells in the S phase, and the G2/M phase cells decreases after drug exposure. Cx43 expression in S and G2/M phase of the cell cycle for cisplatintreated ESCC was significantly altered. Cx43 expression was enhanced in low concentration of cisplatin ( from 0 to 2 μmol/L). In high concentration of cisplatin (from 2 to 12 μmol/L), cellular Cx43 expression was gradually weakened. In particular, Cx43 expression of G2/M phase cells was more active and susceptible to cisplatin.  Conclusions:Our study shows that Cisplatin serves as potential therapeutic target for Cx43 expression of the S and G2/M phase of cancer cell cycle as well as DNA replication of the S phase during cisplatin treating ESCC. The characteristics of cisplatin targeting S and/or G2/M phase of the cell cycle may reduce or avoid  the effects of nondividing cells.

A Study on the Expression and Subcellular Localization of Calcium and  #br# Integrin Binding Protein 1 in 293T Cells
Alignment of CIB1 protein sequence among human, rat and mouse, and study on the expression and subculluar localization of CIB1 in 293T cells. Western blot analysis show that CIB1 protein expression is increased significantly in transfected 293T cells, but undetectable in untransfected 293T cells. To further monitor the expression and subcellular localization of CIB1 in 293T cells, transfections CIB1 exogenous plasmid transfection  with varying time were  detected by immunofluorescence analysis in laser confocal microscope. The results show that  with transfection of CIB1 exogenous plasmid into 293T cells, more and more CIB1 were expressed, and there are obvious translocation of CIB1 from nuclear to cytoplasm. These results will highlight the understanding of CIB1 subculluar localization changes its potential roles.

The Value of Three Immunological Methods in the Diagnosis of Silicosis #br# Complicated with Pulmonary Tuberculosis
Objective:〖WTBZ〗To investigate the value of T lymphocyte ELISA dot experiment (TSPOT), tuberculin skin test (TST) and tuberculosis antibody in the diagnosis of.patients with pulmonary tuberculosis in silicosis. 〖WTHZ〗Methods:〖WTBZ〗Fortyone cases of silicosis TB infection, ninety 〖HJ2.1mm〗cases of non silicosis tuberculosis, forty cases of healthy medical students as control group were collected from August 7, 2015 to May 20, 2016, all the above samples were detected by the above three immunological methods, and the resuts of different methods were analyzed. 〖WTHZ〗Result:〖WTBZ〗1) the positive rates of TSPOT in silicosis, non silicosis tuberculosis and healthy people were 73.17%, 93.33% and 15% respectively, there were significant differences between the two groups in the three methods:The positive rates of TST in silicosis, non silicosistuberculosis and healthy people were 58.54%, 88.89% and 32.5% respectively, compared with non silicosis TB patients, the differences were statistically significant. There were no significant differences between healthy people and silicosis patients, Compared with non silicosis patients, the differences between healthy people and non silicosis patients were statistically significant. The positive rates of TB antibody in silicosis, non silicosis, tuberculosis and healthy people were 36.58%, 42.22% and 52.5%, respectively. There were no statistically significant differences between the two groups in the three methods. 2) the sensitivity of the three detection methods in silicosis patients was 73.17%, 58.54% and 36.58%, respectively, and the differences were statistically significant:The sensitivity of TSPOT and TST was higher than that of TB antibody, the differences were statistically significant, the specificity was 85%, 67.5% and 47.5%, respectively, the differences were statistically significant, The specificity of TSPOT was higher than that of tuberculosis antibody method, the difference was statistically significant; the positive predictive value were 83.33%, 64.86%, 41.67%, and negative predictive value were 75.56%, 61.36%, 64.44%, the differences between positive and negative predictive value of three methods, were statistically significant. 3) the positive rate of TSPOT in I, II, III phase tuberculosilicosis detection was 52.38%, 94.12%, 100%, there were significant difference between the positive rate of I and II phase or III phase, there were no significant difference between II and III positive rate. 〖WTHZ〗Conclusion:〖WTBZ〗TSPOT has high sensitivity and specificity, and is an effective methodin the diagnosis and clinical staging of silicosis.

Toxicological and Safety Assessment of Oligopeptides and Vitamin C Compound in Solid Drink
In order to evaluate the toxicological safety of oligopeptides and vitamin C compound in solid drink. Toxicological and safety assessments were conducted, according to the Technical Standards for Testing and Assessment of Health Food by Healthy Ministry 2003, the results showed that the maximum tolerated dose of oligopeptides and vitamin C compound in solid drink on mice was more than 20 g/kg. BW. Therefore, the sample fell into the scale of nontoxicity. The genotoxicity tests including Ames test,bone marrow cell micronucleus,sperm shape abnormality were all negative. The results of 30days test showed that the animals of control group and each dose group were well developed, without death and abnormal clinical sign. The body weight, food intake  food utilization rate, organ weight, organ / body ratios, hematological parameters and blood biochemical indexes had no significant difference (P>0.05) between the control group and the treatment groups. There are no abnormal pathological changes in liver, kidney, stomach, intestine, spleen,testicle and ovary of rats in treatment group. The conclusion is that the oligopeptides and vitamin C compound in solid drink are safe with no evident toxic effect.

#br# Targeting Effectiveness Analysis of Heart Developmental Candidate Gene TitinCap with #br# Gene Knockout Method CRISPR/Cas9
CRISPR/Cas9 technology, developed rapidly in recent years with a high targeting efficiency, is considered a revolutionary field of genetic technology. TitinCap gene is a candidate gene for zebrafish cardiac development, and there is no knockout strain of zebra fish TitinCap gene in the world so far. In order to study the mechanism of the TitinCap gene in heart development, here the CRISPR/Cas9 gene targeting technique was used to establish the TitinCap gene knockout strain in zebrafish. The sequencing results showed that the embryos injected with CRISPR/Cas9 gRNAs had bimodals, indicating that there was a base deletion or insertion near the target site. The sequencing results demonstrated that the gRNA was effective. In the screened F0 mutants, the sequencing results also showed positive results. All these results indicated that the TitanCap gene of the zebrafish was successfully knocked out with the CRISPR/Cas9 gene targeting technique, and the zebra fish of the TitinCap gene knockout was obtained.

Functional Properties and Volatile Flavor Compounds Composition #br# of Protein Hydrolysates from Grass Carp
The enzymatic hydrolysis of grass carp by protease was carried out to study the functional properties of the hydrolyzate and the volatile flavor compounds of the hydrolyzate. The results showed that the solubilities and thermal stability of the protease and trypsin were better than those of the pepsin hydrolyzate (P<0.05). After enzyme treatment, the percentage of fresh sweet amino acids in the enzymatic hydrolyzate  increased, and the bitter amino acids  decreased. 83 types of volatile compounds were detected by SPMEGCMS, respectively, and aldehydes, ketones and alcohols were the main compounds present. After enzymatic hydrolysis, the relative content of alcohols was reduced, while the aldehyde and ketone content  increased significantly, the hydrolyzate has an unique flavor. After enzymatic hydrolysis of trypsin,proteolytic enzymes and enzymolysis of grass carp can be used as a food base in processing.

#br# Tissue Culture and Rapid Propagation Technology of Tetraploid #br# Barnyardgrass (Echinochloa crusgalli (L.) Beauv)
We report a successful and efficient plant regeneration system using mature embryo of barnyardgrass (Echinochloa crus). The effect of different sterilization procedures on explant culture and the effect of plant growth regulators on regeneration process and acclimatization of transplanting were assessed. It was found that EtOH+0.1%HgCl2 was optimal for explant sterilization. Essentially, dehulled and sterilized seeds were plated on callus induction and subculture medium composed of MS basal salts and DL vitamins plus 2.5 mg/L 2,4D, 300 mg/L glutamine, 〖JP1〗500 mg/L proline, 3% maltose, 0.8% (w/v) agar at pH 5.8. Differentiation was carried out on a differentiation medium MS with 50 mg/L〖JP+1〗 inositol plus 1.0 mg/L 6BA, 0.2 mg/L kinetin KT, 0.5 mg/L NAA, 0.25 mg/L IAA, 3% maltose, 0.8% (w/v) agar at pH 5.8. Plantlets ranging from 3 to 5 cm in length were transferred to the rooting media comprising 1/2MS basal salts. In vitro regenerated plantlets were washed to remove the traces of agar and then were transferred to pots containing nutritious soil and perlite (2〖DK1〗∶1, v/v), and the survival rate was over 90%. The protocol developed in this study will facilitate barnyardgrass generation and is useful for the production of herbicideresistant transgenic plants.
