Abstract:Objective:To study the target effect of cisplatin on treating esophageal squamous cell carcinomas (ESCC). 〖WTHZ〗Methods:〖WTBZ〗In this study, the influence of cisplatin on cell cycle progression of esophageal cancer and connexin 43 (Cx43) expression in cancer cell cycle were detected by bivariate flow cytometric analysis. Results:he effect of cisplatin on cell cycle of ESCC was mainly due to DNA replication of S phase cells, arrest of cells in the S phase, and the G2/M phase cells decreases after drug exposure. Cx43 expression in S and G2/M phase of the cell cycle for cisplatintreated ESCC was significantly altered. Cx43 expression was enhanced in low concentration of cisplatin ( from 0 to 2 μmol/L). In high concentration of cisplatin (from 2 to 12 μmol/L), cellular Cx43 expression was gradually weakened. In particular, Cx43 expression of G2/M phase cells was more active and susceptible to cisplatin. Conclusions:Our study shows that Cisplatin serves as potential therapeutic target for Cx43 expression of the S and G2/M phase of cancer cell cycle as well as DNA replication of the S phase during cisplatin treating ESCC. The characteristics of cisplatin targeting S and/or G2/M phase of the cell cycle may reduce or avoid the effects of nondividing cells.
